It will ONLY tell if a “water-in-oil” emulsion has been generated or not. If the drop stays intact, it is a “water-in-oil water” emulsion. If it “dissolves” quickly, it is an “oil-in-water” emulsion. To test its quality, put a small drop on a glass-slide […]

Can the emulsion be stored? For how long? https://www.bertin-technologies.com/faq/can-the-emulsion-be-stored-for-how-long/ Mon, 25 Oct 2021 14:56:52 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43491 Yes. The emulsion can be stored for up to 4 weeks, depending on the peptide’s stability. Please note that you should NOT keep it in the emulsion tube itself. You should transfer all of the emulsion into the injection syringes, and then store the syringes at 4 °C. The stability of the emulsion has been […]

What type of syringe should I use to recover my emulsion ? https://www.bertin-technologies.com/faq/what-type-of-syringe-should-i-use-to-recover-my-emulsion/ Mon, 25 Oct 2021 14:56:38 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43496 We recommend the use of Braun’s Injekt-F 1 mL syringes. Other 1 mL syringes may be used, at risk of increasing potential leaks and the waste of reagents by augmenting the dead volume at the back of the syringe.

Is the integrity of the peptide/protein compromised by the agitation? https://www.bertin-technologies.com/faq/is-the-integrity-of-the-peptide-protein-compromised-by-the-agitation/ Mon, 25 Oct 2021 14:56:36 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43495 No. The efficiency of the emulsion was tested using MOG 35-55. The results were comparable between emulsions prepared on the same day, 3 weeks before and 15 weeks before immunization. Read more about it in our Application Note.

Is there a risk of the needle coming off while pushing the viscous emulsion? https://www.bertin-technologies.com/faq/is-there-a-risk-of-the-needle-coming-off-while-pushing-the-viscous-emulsion/ Mon, 25 Oct 2021 14:56:33 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43494 To avoid this type of risk, please make sure to attach the needle to the syringe BEFORE pushing in the emulsion.

Has this system been used/validated for generating Collagen II – CFA emulsions specifically? https://www.bertin-technologies.com/faq/has-this-system-been-used-validated-for-generating-collagen-ii-cfa-emulsions-specifically/ Mon, 25 Oct 2021 14:56:31 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43493 Yes. Several customers have validated the use of rat type II collagen and bovine type II collagen in their experiments. The recommended protocol is: Keeping all of your reagents on ice: the tube, the antigen, Freund’s adjuvant and buffers, etc.  Add the reagents to the tube, and shake it for 60 sec. After shaking, put […]

The emulsion has a very runny consistency https://www.bertin-technologies.com/faq/the-emulsion-has-a-very-runny-consistency/ Mon, 25 Oct 2021 14:56:05 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43489 When creating smaller amounts of emulsion (< 4 ml), it can appear as a “runny” emulsion. This can also occur with some antigens (especially some peptides), even when large amounts have been prepared. Importantly, it does not influence the results in any of the disease models we have tested (EAE and CIA).

The emulsion looks like it has NOT been mixed https://www.bertin-technologies.com/faq/the-emulsion-looks-like-it-has-not-been-mixed/ Mon, 25 Oct 2021 14:56:00 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43488 Please check that the receptacle has not been filled up to the top with adjuvant and antigen. If the receptacle is filled up, it will result in NO mixing and an emulsion will not be generated.

What is the minimum amount of emulsion I can prepare with one Precellys Emulsion Kit? https://www.bertin-technologies.com/faq/what-is-the-minimum-amount-of-emulsion-i-can-prepare-with-one-precellys-emulsion-kit/ Mon, 25 Oct 2021 14:55:56 +0000 https://www.bertin-technologies.com/?post_type=faq&p=43487 The minimum amount that we recommend to prepare is 2 ml. Prepare a starting volume of 2.4 ml. Preparing 2 ml to 4 ml generates an emulsion that appears less viscous.  Nonetheless, it still generates a perfect immune response and induces disease in EAE and CIA models.
